人细胞色素P450 2C9 cDNA 的克隆、鉴定及真核细胞表达重组质粒的构建

CLONING AND IDENTIFICATION OF HUMAN CYTOCHROME P450 2C9 cDNA AND CONSTRUCTION OF ITS MAMMALIAN CELL EXPRESSION PLASMID

摘要: 从人肝组织中提取总RNA ,用RT - PCR 方法扩增人细胞色素P450 2C9 的cDNA。以T - A 克隆方法将该基因的cDNA 连接到p GEM - 3Z质粒上,然后通过内切酶图谱分析及部分序列分析加以鉴定,结果显示克隆片段具有人细胞色素P450 2C9 内切酶图谱和核苷酸序列,该片断含5′端和3′端部分非编码区及完整的编码区。最后将该cDNA 片段连接到哺乳动物细胞的表达质粒pREP9 中。为建立表达人细胞色素P4502C9 基因的细胞系打下了基础。

Abstract: With reverse t ranscription polymerase chain reaction (RT2PCR) and DNA recombinant technique , a full2length cDNA encoding the monooxygenase cytochrome P450 2C9 from human liver was cloned into a plasmid p GEM23Z. The cDNA segment was analyzed by rest riction enzyme mapping and DNA sequencing , and was confirmed to be nothing other than CYP 2C9 cDNA. Then the CYP 2C9 cDNA was subcloned into a mammalian expression vector , ready for development of the cell line expressing CYP 2C9.
